GLP-1 initiators lose 1.2 points more weight than SGLT2 starters in All of Us cohort

- 01Retrospective new-user study of 2,932 GLP-1 receptor agonist and 941 SGLT2 inhibitor initiators in the NIH All of Us program found a 12-month weight change difference of -1.20 percentage points (95% CI -1.91 to -0.48), NNT 14.
- 02Effect was -1.96 points in obesity-only patients and absent in type 2 diabetes (+0.08 points, interaction P=.006); women lost 1.71 points more while men showed no benefit; HbA1c fell 0.261% more with GLP-1.
- 03Authors call the interaction findings hypothesis-generating and requiring prospective confirmation; the design is observational with propensity weighting, not randomized, and covers 2018-2023 claims and measurements.
