  1. 01Six RCTs covering 3,459 adults with obesity found the 36 mg maintenance dose of the oral non-peptide GLP-1 agonist orforglipron cut body weight, BMI, HbA1c, waist circumference, triglycerides and systolic blood pressure more than 12 mg.
  2. 02Authors say the findings support the higher maintenance dose when clinically appropriate, a dosing question clinicians will face if orforglipron reaches the market.
  3. 03Small increases in ALT and AST were seen at 36 mg, and the pooled trials were not powered for rare or long-term harms or cardiovascular outcomes.
