GLP-1 use tied to lower amputation risk in diabetic foot ulcer cohort
The association appeared only in patients without coded peripheral arterial disease, where five-year amputation hazard fell to 0.21 versus other glucose-lowering therapy.

The wire
- 01In 21,325 propensity-matched pairs from TriNetX, GLP-1 receptor agonist users with type 2 diabetes and foot ulcer had a five-year lower-extremity amputation hazard ratio of 0.60, and all-cause mortality HR 0.48.
- 02Effect modification was stark: HR 0.21 in the no-PAD stratum versus 0.97 where PAD was coded, a signal clinicians managing diabetic foot ulcers may want to track.
- 03Authors call the retrospective, observational findings hypothesis-generating and requiring prospective confirmation; six pre-specified sensitivity analyses were run, and broadening the PAD definition attenuated the result.
