Systematic review finds tirzepatide raises beta-cell function indices in type 2 diabetes
Eight reports covering 3,442 participants showed HOMA2-B gains of 77% to 163%, but most evidence came from post hoc, industry-funded analyses.

The wire
- 01A PRISMA-guided systematic review in Cureus pooled eight reports across six populations; tirzepatide 5-15 mg weekly raised HOMA2-B 77% to 163% and lifted the clamp disposition index roughly six-fold.
- 02Intact proinsulin fell up to 55% and the proinsulin/C-peptide ratio up to 49%, exceeding placebo, dulaglutide and semaglutide 1 mg, suggesting reduced secretory demand on beta cells.
- 03Heterogeneity prevented meta-analysis, risk of bias was high or serious in three reports, no study assessed function after drug withdrawal, and the review was unfunded and unregistered.
