Lauroyl-L-carnitine outperforms SNAC for intestinal semaglutide absorption in rats
The enhancer boosted uptake through a dynamin-sensitive pathway and raised systemic exposure in beagle dogs and cynomolgus monkeys without overt tissue injury.

The wire
- 01Researchers compared SNAC, N-dodecyl-beta-D-maltoside and lauroyl-L-carnitine for enteric semaglutide delivery; LLC gave the largest absorption increase in rats, an effect substantially attenuated by dynasore co-treatment.
- 02Oral semaglutide currently relies on SNAC for gastric absorption, so an enhancer suited to small-intestinal delivery could change how oral incretin formulations are built.
- 03DDM disrupted tight junctions with reduced barrier recoverability; the work is preclinical across Caco-2 monolayers, rats, dogs and monkeys, with no human dosing reported.
