The wire

  1. 01In adults with type 2 diabetes and overweight or obesity at cardiovascular risk, orforglipron met noninferiority for major adverse cardiovascular events against insulin glargine (HR 0.84, 95% CI 0.59-1.20), reported at EASD and published in The Lancet.
  2. 02The non-peptide oral GLP-1 agonist, already FDA approved for obesity, also showed sustained glycemic and weight improvements over 104 weeks, a 24.2% median drop in urinary albumin-to-creatinine ratio at week 52, and slower eGFR decline.
  3. 03ACHIEVE-4 was not powered for cardiovascular superiority; ATTAIN-Outcomes, testing orforglipron against placebo in established disease, is scheduled to complete in 2031. GI adverse events hit 62.1% versus 14.2% and drove discontinuations.
